Title: I have a problem with connecting the relay (m3/m5) in DDCSV 2.1
I have a problem with connecting the 5v relay, to ddcsv2.1 loletta, I connect it to the M3 / M5 connector, VSO, GND I will add that I want to use the spindle without speed control.
I connect the relay in such a way
VSO ==> Relay VCC
M3 / M5 ==> Relay IN
GND ==> Relay GND
I have a relay with choice of low and high state, I checked both modes without success.
Is it possible to connect such a relay?

The M3/M5 output is isolated from the controller. The coil of your relay should be connected to the positive of an external voltage source to match your relay coil voltage. The negative of the voltage source should be connected to the GND terminal. Your relay coil current should NOT exceed 20ma
See picture 2-13 in the manual
VS0 is the voltage output for PWM control of Spindle Speed.

thanks for the explanation, I connected in this way and works :-)

OK you are not connecting immediately to a relay. The Relay board you have has opto coupling to the relay. To energise the relay the boards Inp needs to be switched to ground, which the controller will do.  I apologise for being pedantic but I would suggest that you do not use the term -5v but preferably 0v. -5v is a voltage 5volts negative to 0v. That is not what you have.
Given your relay board the connections are > PSU 5v connect to VCC and PSU 0v connect to GND. This supplies the 5v for the operation of the relay. Now In1 or 2 connects to the M3/M5 terminal and Controller GND connects to GND. THIS IS NOT WHAT YOU HAVE. It is working but it is incorrect and you should change it.

I've added a bad picture, this one is correct :-)
